چکیده

Rationale The complement anaphylatoxin 5a (C5a) is an inflammatory mediator associated with the pathology of inflammatory diseases such as sepsis. C5a mediates its pro-inflammatory effects via interaction with two C5a receptors, C5aR and C5L2. C5aR initiates G proteincoupled receptor signaling while C5L2 is not coupled to G proteins and its role in C5a-mediated immune processes is not well understood. We hypothesized that mast cells (MC) expressed C5aR and/or C5L2 and that C5a activated MC through these receptors.

The seven-transmembrane receptor, C5L2, is a stimulatory receptor on human mast cells

Background Complement anaphylatoxin 5a (C5a) is a powerful inflammatory mediator involved in the pathology of inflammatory diseases such as chronic idiopathic urticaria. C5a binds to two receptors, C5aR, a G protein-coupled receptor, and C5L2, a 7-transmembrane receptor deficient in G protein coupling. The role of C5L2 in human mast cells (huMC) is unknown. We hypothesized that huMC express C5a...

The interaction between C5a and both C5aR and C5L2 receptors is required for production of G-CSF during acute inflammation.

The complement activation product, C5a, is a key factor for regulation of inflammatory responses. C5a and C5adesArg bind to their receptors, C5aR and C5L2, but the functional roles of C5L2 remain controversial. We screened the patterns of 23 inflammatory mediators in cultures of LPS-activated mouse peritoneal elicited macrophages (PEMs) in the presence or absence of recombinant mouse C5a. Produ...

TLR activation enhances C5a-induced pro-inflammatory responses by negatively modulating the second C5a receptor, C5L2

TLR and complement activation ensures efficient clearance of infection. Previous studies documented synergism between TLRs and the receptor for the pro-inflammatory complement peptide C5a (C5aR/CD88), and regulation of TLR-induced pro-inflammatory responses by C5aR, suggesting crosstalk between TLRs and C5aR. However, it is unclear whether and how TLRs modulate C5a-induced pro-inflammatory resp...

C5a receptor enables participation of mast cells in immune complex arthritis independently of Fcγ receptor modulation.

OBJECTIVE Mast cells are tissue-resident immune sentinels that are implicated in the pathogenesis of inflammatory joint disease. The aim of this study was to test our hypothesis that complement fragments could be key activators of synovial mast cells in autoimmune arthritis. METHODS In vivo studies used the murine K/BxN arthritis model, a distal symmetric polyarthritis mediated by IgG immune ...

Anti-inflammatory Effects of PMX205 in Mouse Macrophage Periodontitis Model

Background: C5areceptor antagonistPMX205 is a synthetic hexapeptidecapable of blocking C5a-C5a receptor (C5aR) axis by simulating C5a active C-terminal amino acid residues. This hexapeptide presents good anti-inflammatory effects in a myriad inflammation models. The anti-inflammatory effect of PMX205 on periodontitis is yet to be fully fathomed. Objective: To examine the anti-inflammatory effec...
